Andy, If you heat up your hotend to print temp you can clean the outside of the nozzle with a small wire brush. Try heating up your hotend to printer temperature. If the temperature is consistent, move the extruder assembly back and forth by hand. If you get those large temperature fluctuations the problem is probably your extruder harness.

Have you checked to see if your 6-pin power connector is fully seated? It is possible to connect and heat your bed and hotend and not have any motor function.
