Did some filament leak out at all?

Next step is to hold the block with a pair of pliers, while it is hot, and push through from the other end with your smallest hex wrench.

You're going to have to be VERY careful not to pull down on the wires and break them while you are pushing the hex wrench through though, so take your time and push gently.

You want to clean it out as much as possible before you put the nozzle back on.

When you take the hotend off and it cools, it's probably going to have some residue still in there. You will hopefully have shifted the blockage out enough to get the rest out with a gentle push.

Or we might try this....heat it up, grab the end that is drizzling down with the tweezers, and gently pull, slowly, as it melts and try to clear that plastic out. It should be fairly easy with an opening that large.

Hang on a sec.....I'll post a pic....

This is the smallest hex wrench - running all the way through a V4 hotend that doesn't have a nozzle on it. It should clear out the barrel almost entirely.

Make sure you have it going straight into the hole - and don't force it in quickly. Gentle slow push.

All you want to do is push out some softened plastic - if you hit a lot of resistance - stop.

Ok let me try again with the hex wrench.

Yes PLA did leak out a bit.

YAAAAAAAAAYYYYYYY! Your barrel is cleared! :D

Okay, let it cool enough to handle it and seat it back in the clamp again. (Still without the nozzle on it.)

You're going to try to feed PLA through the hotend without the nozzle. Should come out in a big-ass glob.

Then retract it from out of the top. Clean any drips off as it cools, and then do two things:

1. If you have a nozzle that you can see through (the new one?) put that one on the hotend.

2. Try to extrude through that nozzle.

Get down at eye level when you seat the clamp and make sure that you have the holes lined up so that the filament is going directly into the top of the hotend. You will be able to see it going in in the gap. Just advance it about 10 mm at a time until you see if it is going into the hotend, or hitting on the side of the hole.

(You can adjust where the hole is by tightening/loosening the clamp a little and you have to be kind of delicate about it.)

Ok, we are working on that...without the nozzle it is coming through just fine. Im letting it cool so I can add the nozzle and try again. I wasn't sure where the holes were you asked me to align but nothing is oozing from anywhere now.

Pull up a chair and sit down where you can see the top of the hotend as it sits in the clamp. The top of the hotend should be just about level with the top of the clamp.

The filament comes out of a hole in the black plastic filament drive above, and then should go straight down into the hole at the top of the hotend. If you get down at eye level you can see it. (It's the only place you can.)

If the holes are out of alignment, the filament will get stuck and not go into the hotend.

If you have any colored PLA other than black you will be able to see it, otherwise it's pretty tough. (working on a photo)

How much did you extrude? It takes at least 50 mm to fill it back up again, but take it in batches of 10 mm.

Here is a picture of green filament going through the holes. (coming out of the filament drive and going into the hotend)

If you get in really close with a flashlight, you should be able to see it from the side.

Unless you have the dual, your filament drive is going to look different than that. Yours should be much easier to see from the left side.
