What is the pupose of the MicroSD card?

Thank you for the above information.

After I initiated the printing from the SD Card and the print job was going, I decided to just exit the Simplify3D program and as soon I did that the print job stopped. I would expect that once I start the print job via SD Card all communication using USB would stop and I should be able to exit the Simplify3D program.

Not if you still have the USB attached. Just disconnect that after the job starts and you can then close S3D.
